dc.description.abstractThe maintenance of gastric mucosal function and integrity highly depends on the status of microcirculation. So, angiogenesis is crucial for gastroduodenal ulcer healing. We studied a novel SNP which was identified at the VEGF promoter region of Korean patients suffering from gastric ulcer. Wild type of VEGF promoter region was cloned and its SNP was generated by site-directed mutagenesis. Through Firefly luciferase assay, the VEGF promoter activity was confirmed to be significantly low in its mutant type in comparison with the wild type in Stomach cancer cell . For in vitro angiogenesis study, HMEC-1 cell transfected with wild type or SNP VEGF promoter driven VEGF expression DNA. Angiogeneic activity was more prominent in HMEC-1 cell expressing wild type VEGF promoter driven VEGF. In conclusion, the novel SNP of VEGF promoter might be the preventive marker of the possible gastric ulceration of Korean.-
