Serum-free culture can minimize the problems that are associating with the serum usage, such as lot-to-lot performance variability and infectious agents. Suspension culture of CHO-DG44 cell which grows anchorage-dependently, is achieved for the industrial-scale production. The CHO-DG44 cells, however, requires the adaptation process to suspension for a loan time. We have developed a serum-free medium suitable to the suspension culture of non-adapted CHO-DG44 cells. This medium was made up on the basis of DMEM/F-12 medium which includes plant hydrolysate, lipid/fatty acid, SITE(Sodium selenite, Insulin, Transferrin, Ethanolamine), trace elements, surfactant, phospholifid. We have achieved 10 passage of culture. Using development media, comparing to the culture with commercial medium, enhanced cell growth and possible long-term culture without adaptation process to suspension. This development medium has the advantage of saving the time of adaptation process as using directly non-adapted cell.-
