dc.description.abstractIt has been recently confirmed that diallyl disulfide (DADS), a garlic derived compound, has antioxidant effect. However, the effect of DADS on neuronal cell has not been investigated, especially focused on the effect upon cell survival signals. Objectives: The effects of DADS on the viability of neuronal cells and cell signals, including phosphatidylinositol 3-kinase (PI3K)/Akt, glycogen synthase kinase-3 (GSK-3) and down stream signals were investigated in PC12 neuronally-differentiated (nPC12) cells by nerve growth factor. Method : To evaluate the toxicity of DADS itself, nPC12 cells were treated with several concentrations of DADS, and 3, (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and trypan blue stain (TBS) were achieved. The levels of free radicals and membrane lipid peroxidation, according to DADS concentration, were evaluated. And, western blotting for the evaluation of the alteration of PI3K/Akt, GSK-3, cytochrome c, caspase-3, and PARP was also achieved when different concentrations of DADS were treated.-
