dc.description.abstract목적: 본 연구는 난소암세포주 SK-OV-3세포에서 마이토콘드리아 단백질인 p32가 pro-apoptotic 단백질인 Harakiri와 관계된 세포사멸에 관한 생리적 기능을 알아보고자 하는데 목적이 있다. 방법: p32와 Harakiri의 immunoprecipitation을 통해 상호작용을 확인하였으며, confocal microscopy를 이용하여 면역형광염색을 통해 세포 내 위치를 확인하였다. 난소암세포주인 SK-OV-3세포에서 p32와 Harakiri를 과발현시켜 36시간 후에 형광현미경에서 GFP가 발현하는 세포를 counting하였다. 결과: p32와 Harakiri는 난소암세포주에서 세포사멸을 일으켰으며, p32와 Harakiri가 세포질 내 마이토콘드리아에 함께 존재함으로써 그 기능을 수행하며 상호작용하고 있음을 알았다. 또한, p32와 Harakiri가 동시에 과발현시켰을 때 세포사멸의 효과가 더 강력함을 알았다. 결론: 본 연구를 통해 난소암세포에서 p32가 새로운 세포사멸에 유도체임을 밝히게 되었으며, p32는 난소암세포 치료제로서 응용도 가능하다고 짐작해 본다.-
dc.description.abstractObjective: The purpose of the study was to examine a possible physiological function of p32-mediated apoptosis signaling in ovarian cancer cells. Methods: SK-OV-3 cells were transfected with respective plasmid DNAs, and cell viability was measured. By immunoprecipitation and immunofluorescence staining analysis, we confirmed that p32 interacts with Harakiri in ovarian cancer cells. Results: In SK-OV-3 cells, p32 interacted with Harakiri and both p32 and Harakiri were colocalized in the mitochondria. In addition, overexpression of p32 induced apoptosis of ovarian cancer cells and augmented Harakiri-mediated apoptosis. Conclusion: Our results demonstrated p32 as an apoptosis inducer and helped to provide the better understanding of the function of p32 in ovarian cancer cells and a possibility of p32 in the application of cancer therapeutics-
