Obesity is caused by the accumulation of excessive lipids at peripheral and adipose tissues. It causes many metabolic diseases such as atherosclerosis. We had developed the vaccine for obesity (named as BRV) treatment using ApoB-100 mimetic peptide. In our previous study, we confirmed that BRV vaccination induced the apoB-100 specific antibody and it blocked the body weight increments for high fat-diet (HFD) fed mice. One of the anti-obesity effects was considered to uptake of LDL with the antibody by macrophages. Furthermore, the lipid uptaken macrophage was free from foam cell formation. In this study, we investigated the fates of VLDL encountered with macrophages in the presence BRV induced antibody. For the metabolic study, quantification of lipids in serum and stool, locomotor activity measurement were analyzed using ICR mice vaccinated with BRV. In results, the BRV induced antibody enhanced the uptake of VLDL by macrophages without foam cell formation. The lipid contents in the stool from vaccinated mice were similar to the normal diet fed group. In conclusion, BRV makes immune complexes with VLDL by recognizing the Apo B-100 and exerted the anti-obesity effects in HFD fed mice.-
