- Abstract
- Statement of Purpose: For past dacades, there has been numerous researches about developing non-viral vector for gene therapy applications. Among them, polymer vectors are attractive, due to its efficient synthesis and modification. This study demonstrates that modification of poly(amidoamine) generation 2 (PAMAM g2) with basic amino acids such as arginine and histidine was evaluated as a gene carrier, in terms of DNA transfection efficiency, cytotoxicity, serum tolerance and endosomal escape. Luciferase assay, MTT assay were performed to confirm the enhanced gene delivery abilities of polyplex. In addition confocal laser microscopy study was performed to visualize the efficient endosomal escape.
