- Abstract
- The development of a simple and rapid technique for detecting programmed cell death-ligand 1 (PDL1) is crucial for cancer diagnosis and therapeutic monitoring. In this study, a novel Quenchbody (Q-body) was developed for rapid and high-throughput one-pot detection of PDL1. Specifically, a new labeling method using both Cystagging and Sortagging was employed to conjugate two dyes of different colors to the anti-PDL1 antibody fragment. The Q-body represented a PDL1 concentration-dependent fluorescence response, and the signal of the hetero-dye-labeled Q-body was enhanced compared with that of a single-dye-labeled Q-body. The anti-PDL1 Q-body generated herein can be used for various applications as a potential diagnostic probe, including screening PDL1 overexpressing cancer cells to facilitate personalized cancer therapy. © 2024 Elsevier B.V.
