Abstract
Objective: to explore how long-term ovarian hormone deprivation for 6 months influence the APP695 expression in the cerebral cortex and hippocampus of Sprague-Dawley rats Design: We extracted the cerebral cortex and hippocampus 6 months after bilateral oophorectomy in 7-8 week-old female Sprague-Dawley rats(n=5) and five control rats. The samples were analysed for APP expression using Western blot analysis with monoclonal antibody to amino acid residue 17-24 of Aβ. The distinction between APP695 and Aβ was made by molecular weight. Results: APP695 expression was decreased in hippocampus and cerebral cortex in oophorectomized rats approximately 77% and 82% respectively that was present in intact rats. That APP695 expressed in brain was 140-KDa which is consistent with mature APP holoprotein. Conclusion: Our results demonstrate that long-term ovarian steroid deprivation induces underexpression of APP695 in the hippocampus and frontal cerebral cortex of female Sprague-Dawley rats. Underexpression of APP695 may be associated with neuronal deterioration which could be associated with decreased cognitive function which is one of early symptoms of Alzheimer disease.
