Abstract
An attempt was made to establish the effects of diethyldithiocarbamate(DDC) which is a copper and metalloenzyme irreversible inhibitor, on the activities of superoxide dismutase(SOD) and catalase in the gastric mucosa of rats. In addition microscopic study of gastric mucosa of rats treated with DDC, 1000mg/kg body weight was carried out. The activities of SOD were analysed by the methods described by McCord and Fridovich(1969) and Crapo et al.(1978), and the activities of catalse was assayed by the methods of Aebi(1983). The results were summarized as follows; 1. At 1 hr. after the administration of DDC, the homogenates of gastric mucosa of rats showed considerably lower activities of Mn-SOD, Cu, Zn-SOD and catalase when compared with those of the control rats (p<0.01). 2. Activities of Mn-SOD, Cu, Zn-SOD and catalase in the gastric mucosa increased sequentially at 1 hr., 3 hr., 7 hr. and 15 hr. after the administration of DDC (p<0.01) 3. Eventually at 7 hr. after the administration of DDC, the activities of Cu, Zn-S0D were similar to those of the control rats. At 3 hr., the activities of Mn-SOD and catalase were similar to the control rats. 4. At 15 hr. after the administration of DDC, the activities of Cu, Zu-SOD and Mn-SOD increased by approximately 2 times its control value, and the activities of catalase increased by approximately 4 times its control. 5. But at 15 hr. after the administration of DDC, the erosive gastritis was present in all of DDC treated rats. At 15 hr. after the administration of saline, the gastric mucosal Iesions of rats were absent. This study confirmed the development of erosive gastritis in rats following the administration of DDC, an copper and metalloenzyme irreversible inhibitor. Therefore superoxide radicals may play an important role in the pathogenesis of gastric mucosal lesion. These results indicate that SOD may be one of the factors in the gastric mucosal defence machanism.
