Fabrication of Terazosin loaded PDLLA Microspheres and the Release Properties

Abstract

Drug delivery system(DDS) is studied so that overcomes a side effect of commercial drugs. One of the purposes is that it extend the duration of drug release. So it is possible to avoid frequent administration of medication. A long term release can be achieved by incorporation of drug in polymeric matrix. A biodegradable polymer was used to the matrix because it is decomposed into harmlessness. So it does not require to remove in the body after complete drug release. There are many biodegradable polymers including poly(amides), poly(amino acids), poly(ester) and poly(urethanes) but the thermoplastic aliphatic poly(esters) such as PLA, PGA and PLGA have been used most frequently due to their excellent biocompatibility and biodegradability. In this study, the polymer matrix used was the PDLLA(an isomer of the PLA) and the drug was the terazosin hydrochloride, a medicine for prostatic hyperplasia. We prepared drug loaded biodegradable polymer for DDS application by two factors those are the particle size and drug content. Drug incorporated biodegradable polymers were prepared using the ultrasonic spray drying method, which is a continuous and easy process, that produces a powder which is non aggregation and high purity. It is confirmed that the products were spherical and up to two micrometers by SEM and DLS. The properties of drug loaded polymer microspheres were characterized by FT-IR. And the drug release was observed by HPLC.
