Abstract
Reactive oxygen species (ROS) generated in cellular metabolism have an effect on ell maturation and develoment. In human reductive tracti ve injury by ROS may indexe female infertility. Als oxidative injury may be responsible for developmental retardation and rest of mammalian preimplantation embryos Activating transcription factor 4 (ATF) is a member of the cyclie-AMP response ekment-binding ICRES family of basic region ine zipper / NTF4 is known to be stress response to protect cell from a stress factors and indeer of pepties. The purpose of this study was to investigate whether ATF4 is involved in the defensive mechanism in orcidative stress condition during the development of m ecimplantation embryo. To verify the extension of ATF4 in dative stress condition, 2-cell stage embryos were cultured in HTF media containing 0.1 MM 0.5 m or mi hydrogen peroxide (HOS for 1hr -cell, 8 hr (4-oil, 17 hr 18-cell, 24 hr wir 18 he cry bastocyst) or 54 be bote bastocyst. The developmental rate decreased in the OlmMH-0. rated group C ed with col group. In embryos treated with 05 ml and I mM HO showed 2-cell block. As a results of the semi-antitative HT-PCR analysis of SODI, ATF4 and Bax gene expression, SODI, ATF4 and Bax gees were increased in 1 ml 05 m, 1 mM HO, treated groups compared with control group in 2-cell embryos, expression of SODI. ATF4 and Bax genes were itably increased in 1 mM, 0.5 mL, 1 mM HO, treated gros cumped with control grow Immunorescence analysis showed that ATF4 rotein was localized at the cytoplasm of preimplantation embryos The increase in ATF4 imm a ctivey was observed in the 0.1 mil05 mm, 1 mM H 0 treated groups compared with control Bro . Il suggests that oxidative stress by H.O. inches expression of ATF4 and may be involved in parte in m anism in preimplantation embryos from xidative injury.
