Abstract
본 연구는 알로에 추출물의 항비만 효능을 검증하기 위해 3T3-L1 지방세포에서의 지방축적 억제효과를 평가하였다. 알로에 추출물 25와 5 0μg/m L 농도에서 모두 DPPH 자유라디칼 소거능이 유의적으로 증가하여 항산화능을 확인했고 원료의 이화학적 특성을 검증하였다. 세포독성을 나타내지 않는 범위 내에서 알로에 추출물의 분화된 지방세포 내 지방축적 억제능을 평가했을때, 각각 8, 12% 유의적으로 지방축적을 억제하였다. 이후 세부적인 지방축적 억제 경로를 확인하기 위해 lipogenesis와 adipogenesis 관련 단백질 발현량을 분자생물학적으로 측정했다. 알로에 추출물은 lipogenesis 관련 단백질 발현에 유의적인 영향을 주지 않았으나, adipogenesis 발생의 주요 전사인자인 C/EBPα와 SREBP1c를 유의적으로 감소시켰다. 따라서 알로에 추출물은 지방세포 내 adipogenesis를 억제하여 지방축적을 억제한 것으로 판단되어 항비만 생리활성 물질로써 가치가 높을 것으로 예상된다.
Obesity is a global health concern that has given rise to many severe diseases such as T2DM, cardiovascular disease, and dyslipidemia. With the increasing prevalence of obesity, the need for anti-obesity bioactive compounds has received increasing attention. Aloe vera Linne is well-known for its wound healing, anti-inflammatory, and anti-fungal properties. To prove the anti-obesity effect of Aloe vera extract (AVE), we estimated the anti-lipogenic effect and antiadipogenic effects of AVE. AVE significantly reduced DPPH radical activity at concentrations of 25 and 50 μg/mL. With all concentrations, AVE significantly inhibited lipid accumulation in 3T3-L1 adipocytes at all the concentrations tested. To elucidate the anti-obesity mechanism, we measured the lipogenic proteins and adipogenic protein levels. AVE did not alter lipogenic proteins, including SCD1, FAS, and ACC1. On the other hand, AVE significantly prohibited adipogenesis by reducing C/EBPα and SREBP1c. To summarize, AVE had an anti-adipogenic effect by modulating C/EBPα and SREBP1c.
