Abstract
The aim of this study was to investigate the inhibitory effects of magnolialide on basophil-mediated allergic reactions and interleukin (IL)-5-mediated B cell growth. Magnolialide was isolated from the leaves of Laurus nobilis L. (Lauraceae). The effects of magnolialide on mast cell stabilization were determined by measuring immunoglobulin (Ig) E-induced β-hexosaminidase activity in rat basophilic leukemia 2H3 (RBL-2H3) cells. Furthermore, IL-4 production, IL-5-induced Y16 pro-B cell proliferation, and the cytotoxic effects of magnolialide on RBL-2H3 cells were studied. Magnolialide inhibited the release of β-hexosaminidase by RBL-2H3 cells. The IC50 value of magnolialide (21.4 M) was lower than the ketotifen positive control (56.2 M). Additionally, magnolialide was able to inhibit IL-4 mRNA transcription and IL-4 production in IgE-induced RBL-2H3 cells with IC50 values of 17.8 μM and 15.2 μM, respectívely. Magnolíalíde also suppressed pro-B cell growth in IL-5-stimulated Y-16 cells in a dose-dependent manner. These results indicate that magnolialide inhibits IgE-sensitized RBL-2H3 cell de-granulation, IL-4 production, and IL-5 stimulated early during B-cell proliferation. These factors are all key parameters involved in the body’s response to a type 1 hypersensitivity reaction.
