- Abstract
- Glutathione S-transferases (GSTs, EC 2.5.1.18) are a family of multi-functional enzymes involved in detoxification of several herbicide classes. A glutathione S-transferases (GSTs) related to the phi (F) and tau (U) class of enzyme only found in plants have been cloned from the Oryza sativa. These genes were expressed in Escherichia coil with the pET vector system and the gene products were purified to homogeneity by GSH- and S-hexyl GSH- Sepharose affinity column chromatography. The ability of the OsGSTF3 and OsGSTU5 to detoxify from the herbicides were investigated by enzyme-catalyed conjugation of GSH with alachlor, metolachlor, atrazine and fluorodifen. The phi class OsGSTF3 displayed highest activity toward acetanilide herbicides, alachlor and metoalachlor. The OsGSTU5 showed the similarity activities toward all herbicides. We propose from these results that the phi and tau class OsGST enzymes show herbicide specificities, and they play an important role in the detoxification reaction of rice.
